Clearance in First Order and Zero Order Elimination Kinetic



  • Clearance is an imaginary volume of plasma from which a drug is completely removed in unit time.


In First-Order Elimination Kinetic

elimination process is not saturated

โ†“

at higher concentrations of drug โ†’ more drug molecules are eliminated &

at lower concentrations of drug โ†’ less drug molecules are eliminated

โ†“

the volume of plasma from which drug is completely removed remains constant

โ†“ i.e. clearance remains constant.


In Zero-Order Elimination Kinetic

โ†“

elimination process is saturated

โ†“

fixed number of drug molecules are eliminated irrespective of plasma concentration of drug

โ†“ at higher concentrations โ†’ less plasma is cleared &

at lower concentrations โ†’ more plasma is cleared

โ†“

i.e. clearance is not constant.
